It's not the fact of the smile, it's the way it was done. That grin would give the Joker the heebee-jeebees.
Seriously, it's just a bit overdone, and takes away a bit from an otherwise excellent sculpt.
I'm assuming Lex is going for a lab/mad scientist look here; I do recall seeing him in a long white lab coat in some old stories from the 50"s. It's not a first appearance thing; Lex originally had a nice full head of red hair (as the link in the post above shows), but the artist on one of his subsequent appearances mistook Lex's bald assistant for the evil mastermind himself, and thus we got the hairless Lex we all know and loathe today...
