Young Frankenstein for sure. The Scary Movie movies didn't do anything for me. They seemed like unfunny attempts at an Airplane! like spoof. I rented The Lost Skeleton of Cadavra from BB online, and it was a pretty good 50's B Movie spoof.
You must try to generate happiness within yourself. If you aren't happy in one place, chances are you won't be happy anyplace. -Ernie Banks
Got to go with Young Frankenstein. Mel Brooks even went through the trouble of filming it in black and white and getting some of the original props from Frankenstein. Have to admire him for that. By the way, does Ed Wood count as a spoof?

Rocky Horror slipped my mind! Good call.
Also a big fan of American Werewolf. Too bad the parisian sequel was so awful.
I think I was one of the few people who loved the spoofery of Gremlins 2 (but I felt the same way about Robocop 2 as well).
Young Frankensteen er Frakenstein, Mad Monster Party, both versions of Invasion of The Sausermen( 1 version the aliens were all covered in eyes, the 2nd they were tiny with ginormous heads), and of course...Beatlejuice>
An American Werewolf in London is number one without a doubt.
Closely followed by Young Frankenstein.
Followed by Return of the Living Dead, The Lost Boys, Fright Night, Freaked, Shaun of the Dead, A & C Meets Frankenstein, Monsters Inc, Monsters House, Gremlins, Re-Animator (yes, it's a comedy) and Evil Dead, (yes, another comedy).
I know a couple of others are escaping me right now.
